Step 3: Isolation of E. coli from certain samples.
Project staff recultured E. coli from water samples with significant levels of E. coli (we chose greater than 64 colony forming units as our minimum) for genetic analysis (ribotyping method) at a later date. To do this, sterile toothpicks were used to select and restreak ten E. coli colonies onto TSA growth medium. The TSA plate was left at room temperature overnight and then refrigerated. Project staff then delivered the isolates to Jackson Estuarine Lab within about two weeks.

Streak
The bacteria are streaked onto a general purpose growth medium (TSA). They will remain viable until they can be transported to Jackson Estuarine Lab.
