Step by Step Overview of Microbial Source Tracking

Step 1: Collect water samples from streams and estuaries.
Step 2: Membrane filtration of water samples for fecal coliform and E. coli.
Step 3: Isolation of E. coli from certain samples.
Step 4: Collect fecal samples.
Step 5: Serial dilution and membrane filtration of fecal samples.
Step 6: Map the results, select which isolates to ribotype.
Step 7: Ribotyping of isolates (contact UNH Jackson Estuarine Lab for more info).
Step 8: Match the unknown-source stream/estuary E. coli to known-source E. coli from fecal samples.
